Efficient Gene Transduction into Mammalian Cells with RetroNectin Reagent

Achieve higher levels of lentiviral transduction and retroviral transduction of mammalian cells

Of all the methods of gene transfer, lentiviral transduction and retroviral transduction are ideal because they allow development of stably transduced mammalian cell lines. RetroNectin® Reagent can dramatically increase the efficiency of lentiviral transduction and retroviral transduction, especially in cells grown in suspension culture (e.g. lymphocytes) and in hematopoietic stem cells. RetroNectin Reagent is a chimeric peptide of recombinant E. coli-expressed human fibronectin fragments with three functional domains: a central cell-binding domain, heparin-binding domain II, and a CS1 site within the alternatively-spliced IIICS region. During the gene transduction procedure, this 574 amino acid protein acts like a matchmaker between recombinant retroviral particles or lentiviral particles and cells, boosting the efficiency of gene transfer – no need to add conventional, cytotoxic additives like polybrene for successful gene transduction. RetroNectin Precoated Dishes are available as 3.5 cm diameter dishes, precoated with RetroNectin Reagent at 40 µg/mL, and ready for use – making workflows for lentiviral transduction or retroviral transduction protocols efficient and simple.

Over 1,900 peer-reviewed articles have been published in which RetroNectin Reagent was used for gene transduction, including studies using human stem cells, primary cells, and difficult-to-transduce cell lines. Excellent results have been obtained with both suspended and adherent mammalian cells, and the levels of gene transfer obtained with RetroNectin Reagent are significantly higher for nearly all cell types when compared to conventional methods with fibronectin, polybrene, or protamine. Additionally, higher efficiencies allow gene transduction to be performed with less virus and fewer cells than conventional methods.

Many studies have used RetroNectin Reagent for gene transduction, including research in the areas of immunology and immunotherapy, gene therapy studies, diabetes research, cancer research, and many studies on hematopoietic stem cells and progenitor cells. While research-grade RetroNectin Reagent (Cat.#s T100A and T100B) are intended for research use only, cGMP-grade RetroNectin is also available. RetroNectin (GMP) (Cat. # T201) is manufactured as a quality-assured product according to guidelines for Good Manufacturing Practice (GMP) for Investigational Products. This product (Cat. #T201) can be used for ex vivo clinical applications and is limited to investigational use only. RetroNectin reagent has been used in over 57 gene therapy clinical trials.
