The BluePrint One Step RT-PCR Kit is designed to perform RT-PCR in a single tube. The mixture of all the components necessary for RT-PCR are prepared in a single tube, therefore it is unnecessary to add reagents in the middle of the process, minimizing the risk of contamination.
A newly developed M-MLV RTase-based reverse transcriptase, BluePrint, is used in the reverse transcription step. This enzyme shows excellent elongation ability for RNA templates containing higher-order structures. For PCR amplification, Takara Bio’s high efficiency, hot start PCR enzyme, TaKaRa Ex Taq HS is used. The BluePrint One Step RT-PCR Kit contains highly optimized One Step BluePrint RT Enzyme Mix, which contains premixed BluePrint RTase, TaKaRa Ex Taq HS, and RNase Inhibitor together with a stabilizing agent; and 2X One Step BluePrint Buffer, a premixed reaction buffer with dNTP mixture, and One Step Enhancer Solution. Use of these premix components makes it extremely easy to prepare the reaction mixture. Combining the Takara Bio RT-PCR technology with a high efficiency hot start PCR enzyme allows this kit to provide a more efficient production of RT-PCR products. Reverse transcription is efficiently performed at 50°C minimizing non-specific amplification. (Higher temperatures can be used because of the One-Step stabilization buffer).
