The 3'-Full RACE Core Set uses a specially designed Oligo dT Adaptor Primer for efficient synthesis from the 3'-end of poly(A) RNA. The Adaptor Primer has 3 restriction sites for easy cloning after RT-PCR. Subsequent amplification uses this adaptor-primer in conjunction with a specific primer for the gene of interest.
Reverse transcription of mRNA followed by PCR is a common method for obtaining cDNA clones for expression analysis. However, it can be difficult to obtain full-length cDNA by this method. Rapid amplification of cDNA ends (RACE) helps overcome this problem by using an oligo dT primer for reverse transcription, followed by amplification with specially designed oligonucleotide primers.
The 3'-Full RACE Core Set uses AMV Reverse Transcriptase XL to maximize the length of the first-strand cDNA, and is recommended for use with Takara's Taq, Ex Taq or LA Taq.
