For certain types of experiments, preventing PCR-induced errors is critical. Such applications include cloning and expression projects, activity studies, structural studies, molecular genetics, and molecular evolution. However, conventional high-fidelity PCR enzymes have limited processivity and limited rate of extension. Their activity is also inhibited if the level of template DNA in the reaction mix is too high. This can frequently lead to reaction failure. Conventional enzymes simply disassociate from the template before synthesis is complete. If there is too much template in the reaction, as can occur when using crude samples, ordinary high-fidelity enzymes will be overwhelmed and, once again, PCR will fail. And conventional high-fidelity enzymes are often incapable of efficient amplification on GC-rich or AT-rich templates or targets with repeated sequences.

To address these situations, Takara's PrimeSTAR GXL and Max DNA Polymerases were developed. These high-fidelity PCR enzymes have a proven track record of performance.

 

PrimeSTAR GXL DNA Polymerase provides high yield, high specificity, and high accuracy for not only standard reactions, but also excels in PCR with GC-rich templates, in the presence of eXcess template, and for amplification of Long products up to 30 kb. It is a workhorse high-performance enzyme that provides high accuracy and unparalleled performance in standard and challenging PCR conditions alike. Rather than spending time subcloning smaller fragments to create constructs with large inserts, you can use PrimeSTAR GXL DNA Polymerase to amplify tens of kb in a single step, with little to no optimization required. Rather than using special enzymes or buffers for "difficult" templates, try PrimeSTAR GXL DNA Polymerase. Studies from both Takara Bio scientists and in the peer-reviewed literature demonstrate its success with templates having >70% GC, >70% AT, and highly repeated sequences.

When fidelity is critical, choose PrimeSTAR Max DNA Polymerase. This unique high-performance DNA polymerase has the highest fidelity and fastest extension speed of any commercially available enzyme, along with extremely high sensitivity, processivity, and specificity. In some cases, extension steps can be as low as 5-10 sec/kb. PrimeSTAR Max DNA Polymerase is also formulated as a premix that supports hot-start PCR, so it's useful for high-throughput studies.

Which enzyme should I use?

PrimeSTAR GXL DNA Polymerase

  • Use on standard and challenging templates alike. Outstanding performance on GC-rich or AT-rich templates and targets containing repeats
  • Samples containing eXcess nucleic acid: tolerates a wide range of template quantity, including high levels of template that inhibit other high-fidelity DNA polymerases
  • Long PCR: amplify products up to 30 kb (human genomic DNA), 40 kb (lambda DNA), or 13.5 kb (human cDNA)
  • Successful, robust, high-yield PCR regardless of conditions
  • Next Generation Sequencing (NGS) studies involving deep sequencing of the same region in many samples

  • Whenever accuracy and fidelity is critical
