Exonuclease III

Exonuclease III is a 3'–>5' exonuclease that is specific for double-stranded DNA or DNA-RNA hybrids. It will degrade double-stranded DNA with a blunt end, a 5'-overhang or a nick, but it will not degrade DNA with a 3'-overhang of at least 4 bases. Exonuclease III is supplied in 25 mM Tris-HCl (pH 8.0), 50 mM KCl, 0.5 mM DTT and 50% glycerol.

Unit Definition

One unit is defined as the amount of enzyme that produces 1 nmol of acid-soluble products in 30 minutes at 37°C and pH 8.0, with restriction-digested calf thymus DNA as a substrate.
