DNA Polymerase I (E. coli)

DNA Polymerase I catalyzes the incorporation of nucleotides into double-stranded DNA in a 5'–>3' direction. It also possesses a 3'–>5' exonuclease (proofreading) activity. The 5'–>3' exonuclease activity is useful for labeling DNA by nick translation. DNA Polymerase I is supplied in 50 mM potassium phosphate (pH 6.5), 1 mM DTT and 50% glycerol.

Unit Definition

One unit is defined as the amount of enzyme that catalyzes the incorporation of 10 nmol of total nucleotides into acid-insoluble products in 30 minutes at 37°C and pH 7.4, using poly d(A-T) as the template-primer.
