Retroviral Vector: pDON-5 DNA

The retroviral vector pDON-5 DNA is intended for high efficiency retrovirus-mediated gene transfer into mammalian cells. Following transfection of the retroviral vector into an appropriate packaging cell line, pDON-5 DNA expresses transient or stably transcribed products containing the psi virus packaging signal, target gene and selection marker. Furthermore, pDON-5 DNA possesses the psi and LTR viral elements but not the structural genes necessary for particle formation and replication (gag, pol, and env). This retroviral vector also contains the human cytomegalovirus (HCMV) immediate early (IE) gene promoter within the U3 region of its 5' LTR; the HCMV IE promoter is a highly active element that enables high retroviral titers. pDON-5 Neo DNA contains a neomycin resistance gene as the drug selection marker for stable transfection.

Purity

  1. Over 70% double-stranded covalently closed circular DNA(RFI) as determined by agarose gel electrophoresis.
  2. Cloning sites confirmed by dideoxy sequencing method.
  3. Single site restriction digest confirmed for PmaC I, Apa I, Sac II, Not I, Bgl II, Cla I, BamH I, Sal I, Hpa I and Xho I (for pDON-5 only).
