Random Primer DNA Labeling Kit

The Random Primer DNA Labeling Kit, Version 2 is designed to yield DNA probes with high activity for hybridization and can be used to label DNA with [32P]-alpha-, [35S]-alpha- or [3H]-alpha-dCTP. This kit is based on a modified method of Feinberg and Vogelstein utilizing random oligonucleotide primers and the cloned exonuclease-free Klenow fragment of E. coli DNA polymerase I. The use of longer 9-mer primers and exonuclease-free enzyme results in higher labeling efficiency and longer probes. This method overcomes many of the disadvantages of the conventional nick translation procedure while producing probes from small amounts of DNA (10 to 20 ng). It can also be used to label DNA fragments that are embedded in low-melting temperature agarose gel slices.

Applications

  • Labeling DNA for use in hybridization probes

Components

Random Primer (9-mer) 60 µL
10X Buffer 75 µL
dNTP Mixture 75 µL
» (0.2 mM each of dATP, dGTP and dTTP)
Exo-free Klenow Fragment (2 U/µL) 30 µL
Control DNA Solution (25 ng/mL) 10 µL
» (Lambda DNA digested with Hind III, 25ng/µL)

Reagents required, but not included in the kit:

  • Sterilized, distilled H20.
  • TE Buffer (10 mm Tris-HCl, pH 8.0, 1 mM EDTA)
  • Labeled dCTP. This kit is designed for use of [alpha-32P]-dCTP (~3000 Ci/mmol); however, [alpha-32S]- and [3H]-dCTP can also be used.
