Cell Surface Glycosphingolipid Degradation: Recombinant Endoglycoceramidase (EGCase II ACT RE)

Recombinant Endoglycoceramidase II (EGCase II) with Activator II (ACT II) is an endo-type enzyme that cleaves the linkages between oligosaccharides and ceramides in acidic and neutral glycosphingolipids, producing intact oligosaccharide and ceramide byproducts. Using EGCase II with protein activator Act II permits cell surface glycosphingolipid degradation in living cells without damage to other cell components.

This enzyme does not hydrolyze cerebroside, sulfatide, sphingomyelin, glycoglycerolipid, glycoprotein and galactosylceramide linkages of gala-type glycosphingolipids. Cell-surface globo-type glycosphingolipids are strongly resistant to hydrolysis by EGCase II ACT RE. EGCase II ACT RE is a mixture of highly purified recombinant EGCase II and Act II.

Storage

Store at or below –20°C until use. Repeated freeze-thaw cycles result in a loss of enzyme activity. Maintain solubilized enzyme at 4°C and use within 2–3 days.

Properties

Optimum pH: 5.5. Enzyme activity reduced to 30% at pH 7-7.5.

  • Inhibitors: 1 mM Hg2+, Zn2+, Cu2+
  • Not inhibited by Ba2+, Ca<2+, Mg2+, EDTA.
  • No inhibition in presence of up to 5% (v/v) fetal calf serum (FCS). 50% inhibition at 10% (v/v) FCS.
