Anti-Human IDH1 Mouse IgG MoAb

Isocitrate dehydrogenases (IDH) catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate, and are divided into NAD(+)-dependent and NADP(+)-dependent classes. Five isocitrate dehydrogenases are currently known. The enzyme encoded by the IDH1 gene in humans is a NADP(+)-dependent isocitrate dehydrogenase found in the cytoplasm and peroxisomes. The IDH1 gene product has been suggested to function in the regeneration of NADPH for intraperoxisomal reduction reactions, and in peroxisomal reactions that consume 2-oxoglutarate. Mutations in the IDH1 gene have been reported in several brain tumors including astrocytoma, oligodendroglioma, glioblastoma multiforme, and secondary glioblastomas. IDH1 is also known as isocitrate dehydrogenase 1 (NADP+), soluble; isocitrate dehydrogenase [NADP] cytoplasmic; NADP-dependent isocitrate dehydrogenase, cytosolic; NADP-dependent isocitrate dehydrogenase, peroxisomal; IDH, IDP, IDCD, IDPC, and PICD.

These products are affinity-purified IgG antibodies that recognize the R132H (Cat.# 10389) and R132S (Cat.# 10391) mutations of human IDH1. The antibodies were raised in rabbit using synthetic peptides, and can be used for Western blot (WB) detection or immunohistochemical (IHC) detection of human IDH1 mutants.

Applications

Recommended antibody concentration (the optimal concentration should be determined empirically):

Lyophilized: stable 5 years at 2–8°C. In solution: stable 2 years at –20°C.

Form

Lyophilized from an antibody solution containing 1% BSA in PBS and 0.05% NaN3. To use, reconstitute in 1.0 mL H2O (final concentration 100 µg/mL).
