TRAP and ALP Double Stain Kit

Bone metabolism is mutually balanced between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. Tartrate-resistant acid phosphatase (TRAP, also known as TRACP) and alkaline phosphatase (ALP) staining enables effective discrimination between osteoblast and osteoclast bone cells. Takara's TRAP and ALP Double Stain Kit combines the chromogenic substrates for alkaline phosphatase, an osteoblast enzyme marker, and tartrate-resistant acid phosphatase, an osteoclast enzyme marker, with a nuclear staining reagent. This enables osteoblast and osteoclast detection due to the simultaneous cell staining for both alkaline and acid phosphatase activities. Detection of both enzyme markers provides a means to study the differentiation of bone cells. The kit substrates are provided as premixed reagents for ease of use. Sufficient reagent is supplied for staining approximately 5 culture plates (24 well format).

An alternative option for the study of bone metabolism is the TRAP and ALP Assay Kit (Cat. #MK301), which contains a soluble substrate that is quickly assayed by absorbance measurement. Either bone metabolism kit may be selected depending on user interest.

Fixation solution
» Citrate buffer (pH 5.4) containing 60%30 mL
acetone and 10% methanol
Sodium tartrate
» 0.5 M sodium tartrate buffer (pH 5.2) 4 mL
Substrate for ACP (premixed)
» NABP/FRVLB 3 vials*
Substrate for ALP (premixed)
» BCIP/NBT3 vials*
Nuclear stain
» Methyl green (in acetic acid solvent)10 mL

*Resuspend the lyophilized substrate in each vial with 10 mL distilled water.
