The accumulation of amyloid-beta protein plaques in the cerebral cortex is a hallmark of Alzheimer's disease (AD). Amyloid-beta is produced by the proteolytic cleavage of amyloid precursor protein (APP), which is mediated by the enzymes beta- and gamma-secretase. This cleavage reaction produces a number of peptides ranging from 36 to 43 amino acids in length. Amyloid-beta insoluble protein is the main component of plaques found in the brains of patients with Alzheimer's disease; however, recent research suggests that soluble, oligomeric forms of amyloid-beta are also involved in this disease.
