Premix WST-1 Cell Proliferation Assay System

The Premix WST-1 Cell Proliferation Assay System enables fast and easy colorimetric analysis of cell proliferation and cell viability in a 96-well plate format. This assay depends on the cleavage of the tetrazolium salt WST-1 to form a dark red, soluble formazan dye product via the action of mitochondrial succinate-tetrazolium reductase found in viable cells. As a cell population increases, so does the amount of succinate-tetrazolium reductase, resulting in a concomitant increase in the conversion of WST-1 to formazan dye. Thus, the quantity of formazan dye is directly proportional to the number of metabolically active cells in cell culture medium. Formazan dye formed by metabolically active cells can be quantified by measuring its absorbance height at 440 nm using a microtiter plate reader.

The WST-1 Assay provides a non-radioactive and spectrophotometric method for determining cell proliferation and cell viability without the need to perform any cell harvesting, washing or solubilization. The assay can also be performed in a single multi-well plate. The Premix WST-1 Cell Proliferation Assay System is supplied as a single, ready-to-use solution containing WST-1 and an electron coupling reagent diluted in sterile phosphate buffered saline. Premix WST-1 should be diluted 1:10 in 100-200 µL cultured cells/well. The WST-1 Assay supplies sufficient reagent for 2,500 assays.

WST-1 and an electron coupling reagent diluted in sterile phosphate
buffered saline.

Storage

Shipped at –20°C. Store at –20°C, protected from light.

If a precipitant is observed during thaw, redissolve it by warming the solution to 37°C for several minutes while gentle stirring. Once thawed, the solution can be stored at 4°C for several weeks (protected from light). For longer storage times, Takara recommends storing the solution in aliquots at –20°C. Do not subject solution to repeated freeze-thaw.

Principle

Tetrazolium salt WST-1 is cleaved to form a soluble formazan dye via the action of succinate-tetrazolium reductase (EC 1.3.99.1), which exists in the mitrochondrial respiratory chain and is active only in viable cells. The measurable activity of mitochondrial succinate-tetrazolium reductase is proportional to the number of live cells. Because increased enzymatic activity leads to an increase in formazan dye production, the quantity of formazan dye is directly related to the number of metabolically active cells in cell culture medium. Furthermore, the formazan dye can be quantitated by reading its absorbance on a plate reader. Formazan dye absorbance levels directly correlate with the number of viable cells in cell culture medium. The Premix WST-1 Cell Proliferation Assay System is designed for the non-radioactive and spectrophotometric quantification of cell growth and viability in proliferation and chemosensitivity assays.
