    Summary: Infectious keratitis is a leading cause of monocular blindness, and can be caused by fungal, bacterial, viral, or Acanthamoeba pathogens. Conventional culture- or microscopy-based assays for keratitis are laborious and may require up to 7 days of culture duration. The authors developed a rapid direct PCR assay using Terra PCR Direct Polymerase Mix and primers for conserved rRNA regions (fungal, bacterial, Acanthamoeba) and the conserved US4 region of herpes simplex virus I. The direct PCR assay showed a total positive detection rate of 82.5% (66/80), significantly higher than culture-based methods. Unlike culture methods that take up to a week, the direct PCR assay only required three hours to generate results.

    Summary: In an effort to develop an assay to detect macrolide-resistant Mycoplasma pneumoniae from crude saliva, a sample type that is easier to obtain from pediatric patients than nasopharyngeal or throat swabs, the authors used a MightyAmp kit (available in Japan; equivalent to Terra PCR Direct Genotyping Kit) for direct real-time PCR. Locked nucleic acid (LNA) probes were developed to discern the A2063G mutation, which occurs in over 90% of macrolide-resistant M. pneumoniae strains in Japan, from wild-type M. pneumoniae. For 51 samples tested, the positive detection rate was 80.4%, lower than assays conducted with extracted DNA; however, the direct real-time PCR assay could be completed in under 60 minutes.

    Summary: In a case study of a 20-year-old male patient with asymmetrical craniofacial and cervical spinal hyperostosis, the authors performed mutational analysis of the oncogene AKT1 in FFPE bone tissue samples. The NucleoSpin FFPE DNA Kit (not available in all geographic locations) and MightyAmp for FFPE kit (available in Japan; equivalent to Terra PCR Direct FFPE Kit) were used for the assay.
