A New Kit for Analysis of Human T-Cell Receptor mRNA Sequences

NGS technology is increasingly being applied to the study of human T-cell receptor (TCR) repertoire diversity. However, common approaches for generating TCR sequencing libraries typically include only portions of TCR variable regions, and often involve complicated multiplex PCR strategies. The SMARTer Human TCR a/b Profiling Kit employs a 5' RACE-like approach to capture complete V(D)J variable regions of TCR-α and/or TCR-β subunits, utilizing a simple, semi-nested PCR strategy for library construction. Libraries produced with the kit are indexed and ready for sequencing on Illumina® platforms. The SMARTer approach:

  • Yields full-length sequence information for variable regions of TCR-α and/or TCR-β subunits
  • Avoids multiplex PCR and minimizes the likelihood of amplification biases during library production
  • Generates high-quality sequencing libraries from 10 ng–3 μg of total RNA or 50–10,000 purified T cells
  • Allows for detection of low-abundance TCR clonotypes, even at shallow sequencing depths

High-Sensitivity RNA-Seq Libraries from Single Cells Captured with the Fluidigm C1 System

The SMART-Seq v4 Ultra Low Input RNA Kit for the Fluidigm C1 System allows for synthesis of high-quality cDNA from IFC-captured cells (96-cell IFC format) processed with the Fluidigm C1 Single-Cell Auto Prep System.
