What are CherryPicker systems?

CherryPicker systems are powerful sample preparation tools for monitoring and capturing a homogeneous population of cells that either express a specific protein or express a specific promoter activity. These systems utilize the chimeric membrane-anchored CherryPicker fluorescent protein, which can be monitored via fluorescence microscopy and captured on magnetic beads via a specific antibody.

Rescue and enrich transfected cells using CherryPicker Systems

  • Separate cells that have been transfected at a low level from untransfected cells

  • Create a homogeneous cell population based on promoter activity or protein expression

  • Capture and enrich your cells in <2 hr using a bench-top protocol, instead of expensive, time-consuming sorting using flow cytometry

How do CherryPicker systems work?

The CherryPicker reporter is only expressed together with your protein of interest (Video 1), or downstream of your promoter of interest (Video 2). Therefore, only the cells with your activity of interest will be enriched. Watch our videos below to see how you can use CherryPicker for both applications.

Enrichment of MEK-expressing cells

  1. HEK-293 cells were transiently transfected with a CherryPicker expression vector encoding a constitutively active mutant of MEK.
  2. Lysates were analyzed for MEK activity by Western blot, using an antibody to phosphorylated MEK substrate, ERK1/2. GAPDH served as a loading control.

Results

  • Before capture: Small fraction of cells express MEK.
  • After capture: Cells with MEK activity are enriched!
