Enrich for Cells of Interest with CherryPicker Systems
CherryPicker cell capture was used to enrich for MEK-expressing cells, as indicated by ERK1 levels (compare the “Before capture” and “After capture” western blot data).
HEK-293 cells were transiently transfected with the CherryPicker-IRES expression vector encoding a constitutively active mutant of MEK (MEK-CA). Transfected cells were mixed 1:10 with untransfected cells to reduce the number of MEK-expressing cells in the population. This mixed cell population was either:
- Immediately subjected to western blot analysis (“Before capture”; Lanes 2–4), or
- Enriched for MEK-expressing cells by performing CherryPicker capture and enrichment before western blotting (“After capture”; Lane 5)
Western blot analysis used a phosphospecific antibody against phosphorylated ERK1. To allow for normalization, the western blot was also tested using an antibody against the ubiquitously expressed GAPDH.
