Label-ON Reagents provide an efficient way to directly label oligonucleotides with various reporter molecules (1–4). All Label-ON Reagents possess the patented 2-aminobutyl-1,3-propanediol (ABPD) backbone, which offers the following distinct labeling advantages:
- Multiple labels can be incorporated because Label-ON Phosphoramidites do not truncate chain elongation at the 5' terminus.
- Natural 3-carbon internucleotide phosphodiester distance is maintained, providing better hybridization and annealing properties than those obtained with other labeling methods.
- Attached label is separated from the phosphodiester connection point to reduce steric hindrance. The ABPD backbone has a 7-atom spacer arm that separates the label from the connection point.
- Label incorporation can be easily measured by standard trityl cation assays because all Label-ON Phosphoramidites possess a dimethoxytrityl (DMT) protected hydroxyl group.
The Uni-Link AminoModifier allows you to directly incorporate a primary amine onto the 5' end of an oligonucleotide using automated synthesis. All Label-ON reagents are provided with a User Manual.
