Label-ON Reagents provide an efficient way to directly label oligonucleotides with various reporter molecules (1–4). All Label-ON Reagents possess the patented 2-aminobutyl-1,3-propanediol (ABPD) backbone, which offers the following distinct labeling advantages:
Multiple labels can be incorporated because Label-ON Phosphoramidites do not truncate chain elongation at the 5' terminus.
Natural 3-carbon internucleotide phosphodiester distance is maintained, providing better hybridization and annealing properties than those obtained with other labeling methods.
Attached label is separated from the phosphodiester connection point to reduce steric hindrance. The ABPD backbone has a 7-atom spacer arm that separates the label from the connection point.
Label incorporation can be easily measured by standard trityl cation assays because all Label-ON Phosphoramidites possess a dimethoxytrityl (DMT) protected hydroxyl group.
