Clontech offers multiple kits for single-cell transcriptome analysis. The core SMART (Switching Mechanism at 5’ End of RNA Template) technology in these legacy kits is referred to as SMART-seq™ in the literature, and has shown to be an immensely powerful tool for transcript counting and studying splice junctions and alternative splice variants in cells. These kits use oligo(dT) priming and allow for cDNA synthesis directly from whole cells, without any additional sample preparation, or from total RNA of high integrity (RIN 8–10). cDNA libraries can be simply and efficiently generated for transcriptome analysis (mRNA-seq) from single cells, a few to 1,000 intact cells, or very low amounts of total RNA (10 pg–10 ng).
The legacy SMARTer Ultra Low kits generate libraries that are compatible with Illumina's HiSeq®, or MiSeq® instruments.
