Tet-One Systems provide all necessary components for tetracycline (tet) inducible expression on a single vector. The Tet-On 3G transactivator is expressed in from the human phosphoglycerate kinase 1 (PGK) promoter, and the cloned gene of interest is expressed from the PTRE3GS promoter. This all-in-one design has been shown to result in up to 25,000-fold induction (Heinz, N. et al., 2011).
There are several options for preparing and delivering Tet-One vectors to mammalian cells. The AAVpro Tet-One Inducible Expression System allows production of high titers of recombinant, adeno-associated virus (AAV) to establish tet-inducible expression of your gene of interest.
