Cellartis Human Pluripotent Stem Cell Services: Reprogramming

With Cellartis reprogramming services, we will deliver high-quality human iPS cell lines from your fibroblast or PBMC samples. The cells will be reprogrammed using footprint-free Sendai virus technology to ensure no viral integration. Features

Our reprogramming package to create iPS cells from your human fibroblasts or PBMCs1 includes:

  • Thawing of your cells, propagation as applicable, and reprogramming using Sendai virus
  • Establishment of iPS cell clones
  • Creation and characterization of Master Cell Banks (QC testing included)

QC testing included with the reprogramming package:

  • Visual inspection and confirmation of the morphologies of the cells in the starting material and in the Master Cell Banks
  • Immunocytochemistry of the cells in the Master Cell Banks to verify the presence of the pluripotency markers OCT-4, SSEA-4, TRA-1-60, and TRA-1-81, and the absence of SSEA-1
  • Sterility testing of the Master Cell Banks (bacteria and Mycoplasma)
  • Confirmation that no Sendai virus vector remains in the cells

Additional QC testing and characterization services for reprogrammed cells:

  • Cellartis Descriptive Karyotyping Service (Y60304)
  • Cellartis Spontaneous In Vitro Differentiation Service (Y60305)
  • Cellartis STR Analysis Service (Y60306)

Details

Cellartis Sendai Virus Reprogramming of Fibroblasts Service (Y60101)

  • Starting materials: 5 vials (1 million cells per vial) of customer-provided fibroblasts, with relevant information regarding culturing and freezing protocols
  • Deliverables: project report, Master Cell Banks of three hiPS clones per donor (5 vials per clone with 2–3 million cells per vial)
  • Lead time: 20 weeks*

Cellartis Sendai Virus Reprogramming of PBMCs Service (Y60102)

  • Starting materials: 5 vials (10 million cells per vial) of customer-provided PBMCs, with relevant information regarding culturing and freezing protocols
  • Deliverables: project report, Master Cell Banks of three hiPS clones per donor (5 vials per clone with 2–3 million cells per vial)
  • Lead time: 18 weeks*

Notes

For safety and QC purposes, we can only work with human samples that are free of human pathogens and that have passed a sterility test. We require documentation proving that all samples shipped to us are sterile and pathogen-free. In the absence of documentation, testing for pathogens (Y60301) and sterility (Y60302) will be performed via a 3rd party service prior to the initiation of Cellartis services. Please note that safety and sterility testing will incur additional lead time and costs.
