Human iPS Cell Culture Protocol Overview

The DEF-CS system is a complete culture system that includes basal medium as well as culture additives and a surface coating compound. Cells are cultured in the absence of feeder cells, streamlining passaging. In addition, cells are passaged as single cells, a method that is less complicated than traditional passaging techniques and is ideal for single-cell applications.

Coating
Coat new cell culture plates with DEF-CS COAT-1a
Media preparation
Mix DEF-CS Basal medium with growth factors (DEF-CS GF-1, GF-2 and GF-3)
Dissociation
Use TrypLE Select Enzyme (Life Technologies) to detach the cells
Cell counting
Add media to the single cell suspension and count the cells
Dilute
Dilute the cells in complete DEF-CS media to achieve a plating density of 4.0–5.0 x 104 cells/cm2
Plating
Remove excess coating substrate and plate the cells
