Reverse transcription of mRNA followed by PCR (RT-PCR) is a common cDNA cloning method; however, it can be difficult to obtain full-length cDNA using this method. Rapid amplification of cDNA ends (RACE) helps overcome this problem with traditional RT-PCR by using an oligo-dT primer for reverse transcription followed by amplification with specifically designed oligonucleotide primers.

The 3'-Full RACE Core Set uses a specifically designed Oligo dT Adaptor Primer for efficient synthesis of cDNA from the 3'-end of poly(A) RNA. The Oligo dT-3 sites Adaptor Primer also encodes three restriction enzyme recognition sites for easy cloning following RT-PCR. Subsequent amplification with this 3' RACE Kit utilizes the Oligo dT Adaptor Primer in conjunction with a specific primer for the gene of interest.

Because the 3'-Full RACE Core Set uses AMV Reverse Transcriptase XL to maximize the length of the first-strand cDNA, it is recommended for use with TaKaRa Taq, TaKaRa Ex Taq or TaKaRa LA Taq polymerases.
