These libraries were constructed in either the pACT, pACT2, or pGAD-GH vectors, which express the GAL4 activation domain fused to high complexity, tissue-specific cDNA, or random peptides. These libraries are compatible with Matchmaker yeast strains Y2HGold, AH109, and Y187; or any previous Matchmaker GAL4 strain.
Traditional libraries are supplied in E. coli, and therefore must be amplified and transformed into yeast before use. For transformations, we recommend the Yeastmaker Yeast Transformation System 2.
Clontech now recommmends using Mate & Plate or Normalized Mate & Plate libraries with Matchmaker Gold.
