High-Throughput Antibody Purification—Capturem Protein A 96

Capturem Protein A 96 is a 96-well plate that provides rapid, high-quality, small-scale purification and screening of monoclonal and polyclonal antibodies. This 96-well plate combines the specific antibody-binding properties of Protein A with novel Capturem technology to provide high-capacity, membrane-based affinity purification in a high-throughput format

Screen your antibodies with a purification method that is:

  • High-throughput: purify 96 antibody samples simultaneously
  • Fast and convenient: room-temperature protocol, complete in under 15 minutes
  • High quality: short residence times reduce the possibility of aggregation or loss of activity
  • High purity: small bed volumes in plate wells trap few contaminants and allow large-volume washes
  • Flexible: purify a wide range of antibodies

Protein A is a cell wall protein from the bacterium Staphylococcus aureus with a high affinity for the Fc region of monoclonal and polyclonal IgG-type antibodies from several different species, e.g., mouse, rat, and human. Capturem kits use a novel high-capacity membrane-based affinity technology that has previously been used to purify highly concentrated his-tagged proteins in just minutes at room temperature, via miniprep columns, maxiprep columns, and 96-well plates.

Capturem Protein A 96-Well Plate

Capturem Protein A 96 consists of a single-use, disposable 96-well plate that yields up to 300 µg of purified antibody per well, depending upon species and antibody isotype. This 96-well plate is suitable for purification of antibodies from animal sera, ascites fluid, cell culture media, and other sources. Capturem Protein A 96 can purify antibodies in under 15 minutes at room temperature, and at a binding capacity of up to 75 mg/ml, far exceeds the capacity of resin-bound Protein A columns. Starting materials, specific antibody affinities, yields, purification times, and binding capacities of individual wells are comparable to individual Capturem Protein A Miniprep columns.

Where resin columns fall short, Capturem Protein A 96 yields highly pure monoclonal and polyclonal antibodies in just 5 minutes, from loading the sample through elution. Short residence times greatly reduce the possibility of antibody aggregation or loss of activity, giving downstream applications a higher probability of success.

Antibody Affinities of Capturem Protein A Miniprep

Species Affinity (µg/column)*

* Whole serum from various species was diluted in a phosphate-based binding buffer (pH 8.0) and bound on Capturem Protein A Miniprep columns. The amount of purified antibody obtained upon elution was determined by measuring absorbance at 280 nm.
