Affymetrix recommends Titanium DNA Amplification Kits for use with its Genome-Wide Human SNP Array 6.0 and 5.0 Sets, as well as with its original GeneChip Human Mapping 250K and 500K Array Sets. These state-of-the-art SNP genotyping systems use Affymetrix’s proprietary GeneChip technologies to interrogate as many as 906,600 SNPs on a single array (in the SNP Array 6.0). Affymetrix evaluated DNA polymerases from several manufacturers and decided to exclusively recommend Clontech’s Titanium Taq DNA Polymerase for genome amplification on the basis of its superior performance and quality.
The Titanium DNA Amplification Kits were designed in collaboration with Affymetrix scientists and come in two formats. The first kit (Cat. 639240) contains enough reagents to perform 300 PCR reactions, while the second kit (Cat. 639243) contains enough for 400 PCR reactions. The 300 reaction kit can be used by itself to support the original two-chip format, enabling you to screen 96 genomic DNA samples with the Mapping 500K Array Set. The 400 reaction kit can be used to support the single chip format, enabling you to screen 48 genomic DNA samples with the SNP Array 5.0 or 6.0 Sets. Robust PCR Polymerase
Titanium DNA Amplification Kits feature Titanium Taq DNA Polymerase, a 5'-exonuclease deficient Taq polymerase blended with TaqStart Antibody for automatic hot start PCR. The Kits also contain an optimized buffer mix (containing MgCl2), purified dNTP mix, and our proprietary RNase-free GC-Melt Reagent, which improves PCR amplification and yield when using templates with high GC-content or complex secondary structure.
Affymetrix GeneChip Technology
All of the Affymetrix GeneChip and Genome-Wide arrays provide high coverage across different populations. The SNP Array 6.0 features more than 1.8 million markers for genetic variation—including more than 906,600 SNPs and more than 946,000 probes for the detection of copy number variation—allowing high-performance, high-powered, low-cost genotyping. The Mapping 500K Array Set is comprised of two arrays, each capable of genotyping an average of 250,000 SNPs, and has been validated in over 120 published studies in less than three years (1). The SNP Array 5.0 features all of the SNPs from the original two-chip Mapping 500K Array Set, as well as 420,000 additional non-polymorphic probes that can measure other genetic differences, such as copy number variation.

Titanium DNA Amplification Kits meet Clontech’s high quality standards and have been functionally validated by Affymetrix scientists for use with their SNP Array 6.0 and 5.0 Sets, as well as the original Mapping 250K and 500K Arrays. This extra validation step ensures that the Titanium amplification step will never be a roadblock to your genotyping success.
