The polymerase in all Titanium products is more robust than other Taq polymerases and allows the amplification of highly complex DNA mixes due to the absence of 5’-exonuclease activity found in wild-type Taq. This novel enzyme contains carefully engineered amino acid substitutions that increase its solubility, making it incredibly sensitive. This SP product performs similarly to standard Titanium Taq DNA Polymerase in all tested customer applications.
Titanium Taq is especially useful in demanding multiplex assays, where protocols are designed to efficiently assess more than one PCR target at a time, using multiple primer sets. This application requires high specificity to avoid mispriming events, as well as the ability to accommodate a variety of Mg2+ concentrations. Titanium Taq is a recombinant polymerase which has been carefully designed for enhanced solubility, resulting in increased sensitivity. Titanium Taq’s tolerance to a wide range of Mg2+ concentration enables more robust performance with targets of variable melting temperatures and amplification efficiencies.

- Amplify targets of up to 2 kb from highly complex templates, such as mammalian genomic DNA. Targets of up to 4 kb can be readily amplified when the template is a moderately abundant cDNA or a less complex genomic DNA
Titanium Taq SP contains the same polymerase and TaqStart Antibody as our standard Titanium Taq products, but with a modified, license-free buffer formulation. This SP product performs similarly to standard Titanium Taq DNA Polymerase in all tested customer applications.
Integrated Hot Start for Higher Specificity
Titanium Taq comes preblended with TaqStart Antibody for higher specificity than other traditional Taq-based methods. This hot start antibody, which is quickly inactivated at the onset of thermal cycling, blocks polymerase activity during the PCR setup, preventing the formation of primer dimers and other artifacts prior to thermal cycling. It is especially useful for multiplex PCR as it limits the number of mispriming events caused by multiple primer sets, which can result in spurious PCR products or smeared gel bands.
