Isolation of Genomic DNA from Blood—Single Preps—NucleoSpin Blood, Blood QuickPure, Blood L/XL & NucleoBond CB
NucleoSpin Blood, Blood L, Blood XL, and Blood QuickPure kits are designed for isolating genomic DNA from whole blood, serum, plasma, buffy coats, and cultured cells in a convenient silica membrane-based spin column format. Maximum starting volumes range from 200 µl of blood for the NucleoSpin Blood and Blood QuickPure mini kits to 2 ml for the Blood L midi kit and 10 ml for the Blood XL maxi kit. The NucleoSpin Blood QuickPure kits are ultra-fast mini kits that include spin columns with a specially treated silica membrane which speeds up the procedure by allowing a combined washing/drying step. This reduces the number of washing and drying steps from three to only one, resulting in a hands-on time of less than 10 min. The DNA purified using any of these kits can be used directly for PCR, Southern blotting, or any kind of enzymatic reaction.
NucleoSpin Blood mini kits only (not the Blood QuickPure & Blood L/XL kits) may also be used for purifying viral DNA for a wide variety of viruses from blood and biological fluids (including HBV, CMV, HPV, TTV, and EBV). The proteinase K step included in the NucleoSpin Blood method removes viral proteins, allowing the viral DNA in the lysate to bind to the silica membrane. Since viral DNA copurifies with cellular DNA, we recommend using cell-free samples (serum or plasma) to prepare pure viral DNA. The purified DNA is suitable for typical downstream applications such as real-time PCR and enzymatic reactions. NucleoBond CBNucleoBond CB kits are designed for the purification of genomic DNA from blood, serum, or cell cultures using NucleoBond AXG anion exchange columns, which yield ultra-pure high molecular weight genomic DNA. Because NucleoBond columns run by gravity flow, shearing is kept to a minimum so that high molecular weight genomic DNA can be easily isolated. The DNA purified by this method is suitable for all kinds of subsequent reactions.
