pKF3 Plasmid

The Enforcement Cloning System pKF3 plasmid allows for easy detection of recombinant DNA clones without the need to screen bacterial colonies. The pKF3 plasmid DNA contains a chloramphenicol resistance gene (CmR) and the lethal gene streptomycin-sensitive ribosomal protein (rpsL) at downstream regions of the tryptophan promoter/operator. This cloning vector is also designed with a multiple cloning site that possesses an amber mutation and 39 restriction sites on the rpsL gene; rpsL will not be expressed if the target gene is inserted into any of these restriction sites. As a result, only targeted clones (containing both the pKF3 plasmid and the target gene) can be grown on LB medium containing chloramphenicol and streptomycin antibiotics.
