Tetracycline-Inducible shRNA- Tet-tTS Cell Lines

Accessory cell lines for Clontech’s Tet-inducible shRNA systems.

Tet-tTS cell lines have been clonally selected to express the Tetracycline transcriptional silencer, or tTS, and greatly simplify the task of creating a tetracycline inducible shRNA cell line, as they are ready-made hosts for our Knockout Tet RNAi Systems H and P.

How tTS Works…

In the absence of doxycycline (Dox), tTS binds tightly to Tet operator sequences (tetO) in the promoter of Clontech’s inducible shRNA vectors. The KRAB domain of tTS then acts as a potent suppressor of shRNA transcription. When Dox is added to the culture medium, tTS dissociates from tetO to allow full promoter activity and high-level shRNA expression.
