Tet-Express Systems—Kit Components

Tet-Express Inducible Expression Systems are a faster, simpler adaptation of the Tet-On and Tet-Off expression systems. Tet-Express requires only a single vector, the Tet-Express transducible protein, and a doxycycline-free protocol. Tet-Express makes tetracycline-controlled transcription a rapid process for all cell types, and is particularly advantageous for cells that are not amenable to sequential rounds of clonal selection.

Tet-Express Plasmid Systems

All plasmid versions of the Tet-Express System contain the following seven components (store all components at –20°C):

  • 10 µg response plasmid (see table below)
  • 25 rxns Tet-Express (includes the Tet-Express Transactivator and Intensifier Reagent)
  • 10 µg control plasmid expressing luciferase under the control of a TRE3G promoter (either pTRE-Luc or pTRE-BI-Luc)
  • 2 µg Linear Hygromycin Marker (also sold separately as Cat. No. 631625)
  • 2 µg Linear Puromycin Marker (also sold separately as Cat. No. 631626)
  • 100 rxns Xfect transfection reagent (also sold separately as Cat. No. 631317)
  • 50 ml Tet System Approved FBS, US Sourced (also sold separately as Cat. No. 631105)

Tet-Express Lentiviral Systems

All lentiviral versions of the Tet-Express System contain the following components. Store all components except the Lenti-X GoStix at –20°C. Store the Lenti-X GoStix at room temperature.

  • 10 µg response vector (see the second table below)
  • 25 rxns Tet-Express (includes the Tet-Express Transactivator and Intensifier Reagent)
  • 10 µg control vector expressing luciferase under the control of a pLVX-TRE3G promoter (pLVX-TRE3G-Luc)
  • Lenti-X HTX Packaging System (Cat. No. 631247)
    • Lenti-X HTX Packaging Mix 2 (Cat. No. 631260; not sold separately)
    • Xfect Transfection Reagent (Cat. No. 631317)
    • Tet System Approved FBS, US-Sourced (Cat. No. 631105)
    • Lenti-X GoStix (Sample)
