Rat cytokine-induced neutrophil chemoattractant-1 (CINC-1) protein is a member of the CXC family of chemokines. The rat CINC family members, including CINC-1, CINC-2-alpha, CINC-2-beta, and CINC-3, are homologous to human GRO/CXCL and mouse MIP-2/KC genes. CINC-1 possesses high sequence similarity to IL-8, and similarly induce leukocyte chemotaxis. CINC-1 also has high similarity to mouse keratinocyte-derived cytokine (KC). Rat CINC-1 is also known as chemokine (C-X-C motif) ligand 1 (CXCL1), GRO-alpha, and melanoma growth stimulating activity alpha (MGSA alpha).
In addition to its established role as a neutrophil chemoattractant, CINC-1 functions as an early acute-phase protein. Levels of CINC-1 in the liver and serum rise in response to brain injury, and CINC-1 is suggested to be responsible for recruitment of neutrophils to the site of injury.
Rat CINC-2 and CINC-3 are also members of the CXC family of chemokines. Rat CINC-2-alpha and CINC-2-beta, differing by only three C-terminal residues, were originally identified as novel neutrophil chemoattractants in conditioned medium of granulation tissue obtained from carrageenan-induced inflammation. Mature CINC-2-beta is a 68-amino acid protein generated by cleavage of a 32-residue signal peptide. CINC-2-alpha and CINC-2-beta have similar chemotactic activity, and they contribute to neutrophil infiltration into inflammatory sites in rats. Other cell types that express CINC-2 proteins include activated macrophages and fibroblasts. Rat CINC-3 is expressed in monocytes and macrophages, endothelial cells, and fibroblasts. CINC-3 interacts with a cell-surface G-protein-coupled receptor known as CXCR2.
