Cadherin Detection

Cadherins, a family of 723–747 amino acid transmembrane glycoproteins that function as calcium-dependent adhesion receptors, play an important role in cell-cell interaction, histogenesis, and cellular transformation. The cadherin family includes E- (epithelial), P- (placental), and N- (neural) cadherin, all of which share a common basic structure but have distinct tissue distributions.

N-cadherin (also known as cadherin 2 (CDH2), CDHN, NCAD, CD325, and CDw325) contains five extracellular cadherin repeats, a transmembrane region, and a highly conserved cytoplasmic tail. It plays an essential role in gastrulation and is required for establishment of left-right asymmetry. The interaction of N-cadherin with intracellular catenin proteins contributes to learning and memory processes.

E-cadherin (also known as uvomorulin, cell-CAM 120/80, and L-CAM) is found on epithelial cells in a variety of embryonic and adult tissues. Studies indicate that unstable or reduced expression of E-cadherin correlates with cancer progression, specifically in cases of lung, breast, and hepatocellular carcinoma as well as gastric and prostate tumors. A soluble 80-kDa form of E-cadherin, presumed to be a degradation product of the intact 120-kDa form, has been reported as elevated in the biological fluids of some cancer patients when compared against healthy individuals.

ELISAs for Cadherin Detection

The Human E-cadherin EIA Kit is a 96-well in vitro enzyme assay is a solid phase E-cadherin (CDH1) ELISA that utilizes two mouse monoclonal E-cadherin antibodies, one of which is coated onto the plate, and the other of which is peroxidase-labeled. It is intended for the quantitative determination of soluble human E-cadherin in serum, urine, cell culture supernatant, and other biological fluids.

Antibodies for Cadherin Detection

Anti-E-cadherin, anti-P-cadherin, and anti-N-cadherin monoclonal antibodies are constructed by fusing immunized host splenocytes with P3-X63-Ag8 myelomas. 

Affinity-purified IgG antibodies that recognize human N-cadherin protein are raised in rabbit using recombinant protein (Cat. # 18571) or synthetic peptide (Cat. # 18573, 18574), and can be used for Western blot (WB) detection, immunohistochemical (IHC) detection, or immunoprecipitation (IP) of human N-cadherin protein.
